I downloaded a single (.iso) file that is just over 4gb in size. How would I be able to transfer this file to my other computer in another room? I have a wireless connection. Will it increase my usage (uploads, d/l's) in any way?

It's best to get a USB. But I doubt you'd be be able to get a 4GB one. So I reckon your best bet would be to use an external hard drive. If you don't want to buy one, then just borrow one from someone.

You can actually get 8 gig USB keys for not much money. One thing you also have to remember is that for FAT drives (iRivers, portable HDD etc) you cannot transfer files over I think 5 gig. In that case you have to split it up using Winrar but I'd recommend that anyway.
