This is all entirely subjective, usually people we would like to be or hang out with, or interesting character studies.

I'll just pick 3 off the top of my head;

Detective Frank Pembleton (Homicide: Life on the Street) - A difficult arseh*le, a terror to face in the box, yet one of the most captivating characters ever on TV. The whole stroke storyline was rather brilliant as well. You are never bored watching Pembleton.

Rorschach (Watchmen) - One of a few intriguing characters from Watchmen, Rorschach is the fan favourite. Basically shows a right wing extreme, and a high code of values. Kind of like Popeye Doyle to the extreme.

Roy Batty (Blade runner) - Just a brilliant creation all round, a perfect specimen, somewhat primal and encapsulates life. He stands out in a dreary, dehumanised futuristic world.

Others of note: Judge Holden (Blood Meridian), Dr Zaius (Planet of the Apes), Noriko (Ozu films), Poppy (Happy Go Lucky), etc.

Also, growing up as a transsexual, I related a lot to Samus Aran in Metroid. A blonde chick wearing a tough suit of armour to get by in a dangerous world. Not a well developed character, but I loved playing as Samus.
 
Mrs. Madrigal and Michael Tolliver from the Tales of the City series of books would definitely be two of my favourite characters ever.
Nobody has or ever will be cooler than Sal Paradise and Dean Moriaty.
I wished Atticus Finch was my dad when I first read To Kill a Mockingbird, and I consider Adrian Mole to be one of literature's great anti-heroes.
If you have kids, they will love Grimble, the eponymous protagonist of the Clement Freud books.

The Martians from HG Wells' War of the Worlds, set a template for alien invasion that's still inspiring (and being plagiarised) 115 years later.

And Yossarian from Catch-22, a lot of fictional characters have made me laugh but none more than him and of course there's a lot more to the character (and story) than just the humour.
 
There's who my imagination deems the greatest then there's the ones who have propelled the actor/actress into something greater..

This guy would make the top region of both lists..

In 2005, Schwarzenegger issued an executive order calling to reduce greenhouse gases to 80 percent below 1990 levels by 2050.[88]
Schwarzenegger signed another executive order on October 17, 2006 allowing California to work with the Northeast's Regional Greenhouse Gas Initiative. They plan to reduce carbon dioxide emissions by issuing a limited amount of carbon credits to each power plant in participating states. Any power plants that exceed emissions for the amount of carbon credits will have to purchase more credits to cover the difference. The plan took effect in 2009.[89] In addition to using his political power to fight global warming, the governor has taken steps at his home to reduce his personal carbon footprint. Schwarzenegger has adapted one of his Hummers to run on hydrogen and another to run on biofuels. He has also installed solar panels to heat his home.[90]

You could argue that if he didn't do "terminator' he'd not of got where he did, he would of been a pot smoking ex body building superstar doing bit parts.
 
This chick is Nancy wake, Cleopatra, Marliyn Monroe, Aung San Suu Kyi, Marie Curie all rolled into one and a bit of robot as well



you could argue that her female nemeises from the same show, is as equally a strong and realistic character..

Actually Rocky Balboa probably deserves a mention as well, for movies one of the most iconic characters created in script and action by the one person.

Sylvester Stallone created the character, wrote the script and his refusal to sell it for what was probably pretty good money for a struggling artist and instead see it through to the end and act the role himself changed his life forever and created a character against which all underdog and sports movies have been measured against ever since.
