How would this show have gone with same character and writing, but as a live action TV Show? Would it still be one of the funniest/best shows of all time? Or would it not have at all the same appeal?

I think there's a 99% chance that the show would have been less funny if it were live-action.
It would be a terrible strain on the animators wrists.

Nah, would have lasted two seasons, the kids growing up would have forced too many changes in the show. Hell, if Bart started off as a 10 year old he'd be pushing 30 by now which would arguably make for worse viewing than what they are currently serving up.
